Why is a black paint adhered on one side of the air-line center conductor of the 16190B?

The black paint is a marking to indicate that the marked side tip of the center conductor should be connected to the test port of the E4991A, 4287A or 42942A. For details, please refer to page 17 of the 16190B operation and service manual.
